Prolactin is a hormone that is mainly synthesized and secreted by lactotroph cells in the anterior pituitary gland. Prolactin signalling occurs via a unique transmembrane prolactin receptor (PRL-R). The structure of the PRL-R has now been elucidated and is similar to that of many biologically fundamental receptors of the class 1 haematopoietic cytokine receptor family such as the growth hormone receptor. The PRL-R is expressed in a wide array of tissues, and a growing number of biological processes continue to be attributed to prolactin [PMID: 30899100].

Poliovirus receptor (PVR, CD155), a member of the nectin-like family of proteins, is recently emerging as a promising target in immunotherapy that enhances the existing anti-tumor responses. Similar to other members of the family, PVR is involved in important cellular processes, such as adhesion, contact inhibition, migration, proliferation, and the immune response [PMID: 18648374]. PVR is dramatically overexpressed in several human malignancies, whereas its expression is low or JPent in most healthy tissues [PMID: 10841575, PMID: 11454801, PMID: 15604290, PMID: 15536144, PMID: 19801517]. Consistent with PVR biology, its overexpression promotes tumor cell invasion, migration, and proliferation and is associated with a poor prognosis and enhanced tumor progression [PMID: 16322240, PMID: 17893876, PMID: 25743219, PMID: 25862891]. Simultaneously, PVR has a considerable immu noregulatory potential. It interacts with the (co)stimulatory receptor DNAM-1 and the inhibitory receptors TIGIT and CD96, resulting in either immune cell activation or inhibition, respec tively [PMID: 28870470].

Mesothelin/MSLN can be proteolytically cleaved into the following two chains by a furin-like convertase: Megakaryocyte-potentiating factor (MPF) and the cleaved form of mesothelin. Both MPF and the cleaved form of mesothelin are N-glycosylated. Mesothelin/MSLN can interacts with MUC16. The membrane-anchored form of MSLN may play a role in cellular adhesion. MPF potentiates megakaryocyte colony formation in vitro.

Human TNFSF10 is a type II transmembrane protein with an intracellular N-terminus and a ‘TNF homology domain’ (THD) at the extracellular C terminus. TNFSF10 can interact with several distinct receptors. Two of these receptors that belongs to TNFR superfamily, DR4 (TRAIL-R1) and DR5 (TRAIL-R2/TRICK2), are plasma membrane proteins containing intracellular death domains essential for activating apoptosis. TNFSF10 is promising for cancer therapy because it is cytotoxic and activates apoptosis in the majority of malignant cells, but not in normal cells.

N-terminal propeptide of type 1 precollagen (P1NP) is one of the markers of bone formation. Osteoblasts contain a large amount of typeⅠprecollagen. During bone formation, typeⅠprocollagen is secreted outside the cell and cleaved by specific protease to form typeⅠcollagen N-terminal procollagen of type 1 precollagen (P1NP) and C-terminal procollagen of type 1 precollagen, PICP) and typeⅠcollagen 3 fragments. Type I collagen is assembled in osteoid where inorganic minerals (calcium and phosphorus) are deposited to form hydroxyapatite (osteoid mineralization). P1NP and PICP enter the blood and urine as metabolites, so the detection of P1NP and PICP can reflect the level of bone formation. As the research evidence of P1NP reflecting bone formation is richer than PICP, P1NP is a better marker of bone formation. P1NP has good specificity, little influence on circadian rhythm and feeding, and is stable at room temperature. At present, P1NP is generally recommended as the preferred bone formation marker.
