稳定同位素标记(SIL)的蛋白和治疗性单克隆抗体

稳定同位素标记(SIL) 的分析物蛋白质被广泛认为是使用LC-MS绝对定量蛋白质的最佳内标(IS )，SIL蛋白经常被认为是内标的’金标准’。

使用SIL蛋白作为内标的好处是与目标蛋白分析物具有相同的理化特性。这意味着可以在样品制备工作流程开始时引入内标，并在整个分析过程中与分析物蛋白一起进行处理(见下图)处理流程。在样品提取开始时添加SIL蛋白可以控制由于免疫亲和或其他分离技术、酶消化、其他分析前处理以及LC和质谱电离引起的变化。
