■ 制品说明
本制品是蛋白质样品进行PAGE(聚丙烯酰胺凝胶)电泳用的上样Buffer。制品中含有示踪色素溴酚蓝(Bromophenol Blue)，可以确认电泳的进行情况。使用本制品，蛋白质在电泳胶中可以得到很好地分离。电泳后用考马斯亮蓝或者银染色等方法染色后，蛋白质电泳带能够得到清晰显示。蛋白质的迁移速度与其固有的分子量、分子的形状及所带的电荷量等有一定关系。
 
■ 保存
-20℃。
 
■ 纯度标准
无蛋白质分解酶杂质污染。
本制品2 μl与1 μg的BSA和AcBSA (乙酰化的BSA) 混合物在37℃条件下保温4小时后电泳，蛋白质的电泳谱带不发生变化。
 
■ 使用方法
向6 μl的蛋白质样品中加入本制品2 μl(本制品与蛋白质样品按照1：3的体积比混合)，振荡混匀后稍离心，即可上样电泳。
本制品使用方便，可以根据蛋白质样品和电泳胶孔的体积，按照上述比例调整本制品的使用量。
 
■ 注意事项
1. 溶解后请按照使用情况适当分装保存，以避免污染及劣化。
2. 本制品可以于-20℃长期保存；室温放置3个月可以正常使用。

概述

Protein A 和 Protein G 磁珠是一种能用于少量分离纯化免疫球蛋白的亲和基质。它们对来源于多种哺乳动物种属的 IgG 亚型具有较高亲和力，如：人、兔和小鼠。本品是将重组的截短型 Protein A 和 Protein G 共价耦联到一种无孔的顺磁颗粒上而形成的。截短型蛋白展示了对 IgG Fc 区更高的单位结合力和降低的非特异结合力。Protein A 和Protein G 对各种 IgG 的亲和性随物种不同和同种属 IgG 亚型不同而不同（见表）。

由于 Protein A 和 Protein G 与磁珠的耦联在较宽的 pH 范围内都是稳定且致密的，因此可以对来自腹水、血清或细胞培养上清物中的 IgG 进行磁性免疫纯化，基质随后还可以进行再生而不影响结合容量。它们还可用于免疫沉淀法，用所选一抗将目的蛋白从细胞粗提液中沉淀。此外，利用 Protein A 或 Protein G 磁珠还能制成可重复使用的免疫共沉淀磁珠。特定的抗体通过化学键交联到被 Protein A 或 Protein G 包被的磁珠表面，形成一个可重复使用的免疫共沉淀磁珠，避免了抗体与目的抗原共同洗脱。 
 
该磁珠颗粒贮存于 1 ml 含 0.1% BSA，0.05% Tween-20和 0.05% NaN3 的 PBS (pH 7.4) 缓冲液中。

Human Club cell protein（CC16、CC10、子宫珠蛋白、尿蛋白1或克拉拉细胞分泌）是分泌珠蛋白家族的成员，是非连接的分泌产物。细支气管俱乐部细胞。它的功能还有待充分阐明，但令人信服的数据表明其作为免疫调节剂和抗炎剂的作用。Club cell protein抑制磷脂酶A2活性以及干扰素γ信号传导和Th1与Th2淋巴细胞调节。血清、血浆和支气管肺泡灌洗液中的俱乐部细胞蛋白浓度测定。自1994年以来在众多研究中使用的洗液。在血清中，它的增加与年龄和石棉、氯化氮和臭氧暴露。更高水平的CC16在结节病、肺纤维化和高PEEP通气患者。血清减少在吸烟者和慢性病患者的肺切除术后发现CC16水平。阻塞性肺病、哮喘或二氧化硅暴露。患此病的胎儿羊水中也发现CC16浓度降低。各种机制引起的肺发育不良（膈疝、糖尿病）嗜好、特纳和唐氏综合征）。在胸腔积液中，CC16浓度出现。与肺中CC16高水平的扩散有关。胸膜充血。基于以上报道，Club cell protein可能是有前景的有用诊断标志物。肺部疾病和损伤。

Interleukin-6 (IL-6) is a pro-inflammatory cytokine that also has an important role in immunity. Mouse IL-6 appears to be directly involved in the responses that occur after infection and injury and may prove to be as important as IL-1 in regulating the acute phase response. Mouse IL-6 is reported to be produced by fibroblasts, activated T cells, activated monocytes or macrophages, and endothelial cells. It acts upon a variety of cells, including fibroblasts, myeloid progenitor cells, T cells, B cells and hepatocytes. IL-6 has a wide variety of biological functions: it plays an essential role in the final differentiation of B-cells into Ig-secreting cells, it induces myeloma and plasmacytoma growth, nerve cells differentiation in hepatocytes, and acute phase reactants.
