BioXcell InVivoPlus anti-mouse PD-1 RMP1-14单克隆抗体与小鼠PD-1(也称为CD279)反应。PD-1是一种50-55kDa的细胞表面受体,由Pdcd1基因编码,属于Ig超家族的CD28家族。PD-1在CD4和CD8胸腺细胞以及活化的T和B淋巴细胞和髓细胞上瞬时表达,在成功消除抗原后PD-1的表达下降。此外,在B细胞前阶段,Pdcd1 mRNA在发育中的B淋巴细胞中表达。PD-1的结构包括ITIM(基于免疫受体酪氨酸的抑制基序),这表明PD-1负调控TCR信号。PD-1通过结合B7家族的成员PD-L1和PD-L2发出信号。在配体结合后,PD-1信号传导抑制T细胞活化,导致增殖减少,细胞因子产生和T细胞死亡。此外,PD-1敲除动物表现出扩张型心肌病、脾肿大和外周耐受丧失,在小鼠的外周耐受性和预防自身免疫性疾病中发挥关键作用。诱导的PD-L1表达常见于许多肿瘤,包括鳞状细胞癌、结肠癌和乳腺癌。PD-L1过度表达会导致肿瘤细胞对CD8T细胞介导的裂解的抗性增加。在黑素瘤的小鼠模型中,可以通过用阻断PD-L1与其受体PD-1之间相互作用的抗体治疗来暂时抑制肿瘤生长。由于这些原因,目前正在探索抗PD-1介导的免疫疗法作为癌症治疗。与J43抗体一样,RMP1-14抗体已显示阻断小鼠PD-L1-Ig和小鼠PD-L2-Ig与PD-1的结合。

BioXcell InVivoPlus anti-mouse CTLA-4 (CD152)克隆号9D9单克隆抗体能与小鼠CTLA-4(细胞毒性T淋巴细胞抗原-4)反应,CTLA-4也称为CD152。CTLA-4是一种33 kDa的细胞表面受体,由属于免疫球蛋白超家族CD28家族的Ctla4基因编码。CTLA-4在活化的T淋巴细胞和B淋巴细胞上表达。CTLA-4在结构上类似于T细胞共刺激蛋白CD28,两种分子都与B7家族成员B7-1 (CD80)和B7-2 (CD86)结合。在与配体结合时,CTLA-4负调节细胞介导的免疫反应。CTLA-4在诱导和/或维持免疫耐受、胸腺细胞发育和保护性免疫调节中起作用。CTLA-4在免疫下调中的关键作用已经在CTLA-4缺陷小鼠中得到证实,这些小鼠在3-5周龄时由于淋巴增生性疾病的发展而死亡。CTLA-4是通过免疫检查点阻断作为癌症治疗靶点的抑制性受体之一。

BioXcell InVivoPlus anti-mouse PD-L1 10F.9G2单克隆抗体与小鼠PD-L1(也称为B7-H1或CD274)反应。PD-L1是属于Ig超家族的B7家族的I型跨膜蛋白,分子量为40kDa。PD-L1在T淋巴细胞、B淋巴细胞、NK细胞、树突状细胞以及IFNγ刺激的单核细胞、上皮细胞和内皮细胞上表达。PD-L1与CD4和CD8胸腺细胞以及活化的T和B淋巴细胞和骨髓细胞上的受体PD-1结合。PD-L1与PD-1的结合导致抑制TCR介导的T细胞增殖和细胞因子产生。PD-L1被认为在肿瘤免疫逃逸中起着重要作用。诱导的PD-L1表达在许多肿瘤中很常见,并导致肿瘤细胞对CD8 T细胞介导的裂解的抗性增加。在黑色素瘤的小鼠模型中,可以通过用阻断PD-L1和PD-1之间相互作用的抗体进行治疗,暂时抑制肿瘤生长。10F.9G2抗体已被证明可以阻断PD-L1和PD-1之间以及PD-L1和B7-1之间的相互作用(CD80)。

BioXcell InVivoPlus anti-mouse CD8α克隆号YTS 169.4单克隆抗体能与小鼠CD8α反应。CD8抗原是一种跨膜糖蛋白,作为T细胞受体的共受体。与TCR一样,CD8与抗原呈递细胞(APC)展示的I类MHC分子结合。CD8主要在细胞毒性T细胞表面表达,但也可在胸腺细胞、自然杀伤细胞和一些树突状细胞亚群中发现。CD8最常见的存在形式是由一个CD8α和一个CD8β链组成的异二聚体,然而,它也可以是由两个CD8α链组成的同二聚体。CD8α和CD8β链均与免疫球蛋白可变轻链具有显著同源性。每个CD8链的分子量约为34 kDa。BioXcell InVivoPlus anti-mouse CD8α克隆号YTS 169.4抗体在体内使用时显示出depleting活性。

BioXcell InVivoPlus anti-mouse Ly6G 1A8单克隆抗体与小鼠Ly6G反应。Ly6G分子量为21-25kDa,是GPI锚定的细胞表面蛋白Ly-6超家族的成员,在细胞信号传导和细胞粘附中发挥作用。Ly6G在发育过程中由骨髓谱系中的细胞(包括单核细胞、巨噬细胞、粒细胞和嗜中性粒细胞)差异表达。单核细胞通常在发育过程中瞬时表达Ly6G,而成熟的粒细胞和外周嗜中性粒细胞保持表达,使Ly6G成为这些群体的良好细胞表面标志物。与BioXcell RB6-8C5抗体不同,1A8抗体与小鼠Ly6G特异性反应,而与Ly6C没有交叉反应性的报道。

克隆号1D11.16.8单克隆抗体能与小鼠、人、大鼠、猴、仓鼠、犬和牛TGF-β(转化生长因子β)亚型1、2和3反应。TGF-β是一种多功能细胞因子,调节上皮细胞、内皮细胞、成纤维细胞、神经元、包括T淋巴细胞和NK细胞在内的淋巴细胞以及其他造血细胞类型的增殖。TGF-β还调节活化巨噬细胞的活性和调节性T细胞的发育。此外,TGF-β在免疫功能、组织重塑和伤口修复中发挥作用。TGF-β以五种高度相似的亚型(TGF-β 1-5)存在,同源性为70-80%。TGF-β1是通过酶切由TGFB1基因编码的长前体TGF-β1多肽合成的,该多肽产生成熟蛋白和潜伏期相关肽(LAP)。LAP和成熟的TGF-β1在分泌过程中非共价结合。TGF-β在许多细胞类型中普遍表达,包括巨噬细胞和血小板,它们表达高水平的TGF-β。TGF-β信号已被证明在癌症、自身免疫性疾病、哮喘、心脏病和糖尿病中发挥作用。TGF-β基因敲除小鼠表现出造血和内皮分化缺陷,并死于严重炎症,这说明了它的重要性。克隆号1D11.16.8单克隆抗体是一种中和抗体。
