图1.常规IgG与VHH的比较,驼科动物如羊驼只产生重链IgG。IgG2和3的抗原结合结构域(Fab片段)不包含重链的CH1结构域,并且轻链也不存在。

 

Jackson ImmunoResearch纳米二抗是羊驼产生的多克隆VHH片段抗体(纳米抗体)。它们对人、兔或小鼠具有特异性。小于15kDa的VHH片段抗体比传统的全IgG抗体小10倍,非常适合需要良好穿透性的成像实验。纳米二抗经过交叉吸附处理,对目标物种具有高度特异性,与其他常用物种的交叉反应性极低,使其适用于多种标记实验。它们可与报告酶和一系列荧光染料结合,包括Alexa Fluor,为高分辨率免疫组织化学和免疫荧光提供了选择。


纳米二抗的优势:

1、小尺寸可以获得更高分辨率的图像

纳米抗体的大小是常规抗体的几分之一,很容易被用作共聚焦和高分辨率成像的工具,纳米抗体直径小,是用于间接标记的常规抗体复合物大小的五分之一(24-30nm),减少了探针和抗原之间的距离,允许捕捉更高分辨率的图像。

2、多克隆保证可靠和优越的信号

3、卓越的穿透力和清除能力

纳米抗体由于其较小的尺寸而表现出优异的组织穿透性,并且由于其扩散系数低于常规抗体,这使其能够在组织中自由移动。缺乏可结晶片段(Fc)进一步提高了该分子从组织中的清除能力,使其成为免疫PET等诊断成像技术的优秀工具。

4、适用于死细胞或活细胞样本染色

具有Fc片段的常规免疫球蛋白通常仅适用于固定样品的染色,因为活细胞的受体会通过Fc片段结合影响生物功能。所以缺少Fc片段的纳米抗体可用于活细胞的免疫染色。

 

5、交叉吸附保证获得更好的特异性和更低的背景
