9D9-CP006单克隆抗体是原始9D9单克隆抗体的重组嵌合型抗体。可变结构域序列与原始9D9克隆号相同,但是恒定区序列已经从小鼠IgG2b变为小鼠IgG1。9D9-CP006单克隆抗体能与小鼠CTLA-4(细胞毒性T淋巴细胞抗原-4)反应,CTLA-4也称为CD152。CTLA-4是一种33 kDa的细胞表面受体,由属于免疫球蛋白超家族CD28家族的Ctla4基因编码。CTLA-4在活化的T淋巴细胞和B淋巴细胞上表达。CTLA-4在结构上类似于T细胞共刺激蛋白CD28,两种分子都与B7家族成员B7-1 (CD80)和B7-2 (CD86)结合。在与配体结合时,CTLA-4负调节细胞介导的免疫反应。CTLA-4在诱导和/或维持免疫耐受、胸腺细胞发育和保护性免疫调节中起作用。CTLA-4在免疫下调中的关键作用已经在CTLA-4缺陷小鼠中得到证实,这些小鼠在3-5周龄时由于淋巴增生性疾病的发展而死亡。CTLA-4是目前肿瘤免疫检查点治疗的热门靶点之一。

带您了解免疫检查点靶点之CTLA-4

在全球范围内,癌症仍然是死亡和发病的主要原因。早期诊断和癌症治疗的进步提高了癌症患者的存活率,但在这些患者中有相当一部分表现出耐药性、转移和复发。在癌症治疗的困境和挑战中,一个有希望的治疗方式就是免疫疗法,利用T细胞群直接杀死恶性肿瘤细胞,其中一个热门靶点就是CTLA-4。


CTLA-4名称为细胞毒性T淋巴细胞抗原4 ,在Treg细胞群上表达的一组免疫调节受体。多篇文献研究表明CTLA-4的免疫检查点阻断提高了肾细胞癌、黑色素瘤、非小细胞肺癌(NSCLC)和头颈部鳞状细胞癌的存活率。CTLA-4是T细胞共刺激配体CD80/86的第二个受体,其功能对下调免疫反应至关重要。与抗原依赖性的第一受体(CD28)相反,CTLA-4是抗原非依赖性的。2011年,Ipilimumab是第一个获得FDA批准用于治疗晚期黑色素瘤的针对CTLA-4的免疫治疗药物,与标准疗法相比,该抗体药物提高了患者的存活率。


T细胞受体(TCR)与通过主要组织相容性复合体(MHC)呈递的抗原的结合需要通过CD28传递的共刺激第二信号来激活。CTLA-4是一种竞争性CD28同源物,结合CD28配体CD80/86,阻止T细胞活化。CTLA-4也介导CD28配体CD80/86的胞吞作用。PD-1与其配体PD-L1结合,负调节T细胞活化。CD28也是PD-1的第二靶位,也是两条通路的交汇点。TCR激活后,CTLA-4和PD-1表达均上调。这两种途径的细胞内信号是通过抑制PI3K下游信号的含有蛋白酪氨酸磷酸酶(SHP-2)的磷酸酶Src同源区-2介导的。此外,CTLA-4与丝氨酸/苏氨酸磷酸酶PP2A相互作用,使AKT去磷酸化,进一步抑制该途径。
