TL2-C29 是一种Toll样受体2 (TLR2)的小分子抑制剂 [1],TLR2在检测来自细菌、真菌、寄生虫和病毒的多种病原相关分子模式(PAMPs)中起着非常重要的作用 [2]。TLR1和TLR6的共受体(co-receptors)对TLR2 信号和配体的特异性有关键影响。细胞表面的TLR2/TLR1和TLR2/TLR6异二聚体可分别结合三酰化和二酰化脂蛋白 [3,4]。TLR2信号是由配体诱导的TLR2异二聚体和衔接蛋白(adapter proteins)的基本细胞质TIR(Toll/IL-1受体)结构域的二聚化启动。随后的信号级联导致促炎症反应。
