支原体检测试剂盒 PlasmoTest™
PlasmoTest™提供一种简便、快速、可靠的可视化方法检测细胞培养中的支原体污染。这种方法首次使用细胞检测支原体。
PlasmoTest™包括两种主要成分:支原体感受器表达细胞和HEK-Blue™ Detection检测培养基。
支原体感受器表达细胞可以通过HEK-Blue™ Detection培养基颜色变化检测支原体。支原体感受器细胞是通过一病原体受体—Toll 样受体2(TLR2)来检测支原体。在支原体存在的情况下TLR2启动下游信号通路,活化NF-kB和一些其他的转录因子。这些转录因子诱导SEAP(分泌型胚胎碱性磷酸酶)分泌到上清中,SEAP可以引起HEK-Blue™ Detection培养基的蓝紫色变化。

内容摘要
HEK-Blue™-2 细胞是支原体感受器细胞,由HEK293改造获得。细胞稳定表达TLR2及活化其信号通路所需的多种基因,同时共表达优化的 SEAP 报告基因。SEAP 报告基因受控于转录因子NF-kB 和 AP-1启动子。
HEK-Blue™ Selection包含多种基因筛选抗生素,这些抗生素保证HEK-Blue™-2 细胞中各种转基因的稳定性。 此外,试剂盒含有Normocin™ ,用以保护HEK-Blue™-2 细胞免受支原体,细菌或真菌的污染。
HEK-Blue™ Detection是特异性检测SEAP的试剂。在SEAP水解后,底物即可出现紫色或蓝色的变化。
