白三烯B4酶免检测试剂盒(Leukotriene B4 EIA Kit)

花生四烯酸是细胞膜的主要组分之一, 通过环氧合酶(cyclooxygenase, COX)和 脂 氧 合 酶 (lipoxygenase, LOX)途 径分别产生以前列腺素E2(prostaglandin E2, PGE2)和白三烯B4(leukotriene B4, LTB4)为代表的类花生酸类物质。LTB4由响应炎症介质的白细胞产生, 并活化多种白细胞功能, 包括白细胞聚集、离子流活化、增强溶酶体中酶的释放、超氧阴离子的产生、趋化等。白细胞活化后,血浆中LTB4的水平从<100pg>100ng/ml。LTB4在白细胞和肝细胞中代谢为20-羟基LTB4和20-羧基LTB4, 不排泄在尿液。

半胱氨酰白三烯 (Cysteinyl leukotrienes, CysLTs)检测试剂盒

半胱氨酰白三烯(Cysteinyl leukotrienes, CysLTs)是花生四烯酸经5-脂氧合酶催化途径生成的代谢产物。花生四烯酸经过5-脂氧合酶催化形成中间产物白三烯A4(LTA4), LTA4再经过两条代谢途径形成LTB4和LTC4, LTC4经酶解经一步转化为LTD4和LTE4。LTC4, LTD4, LTE4在第六位上均有半胱氨酸, 故称半胱氨酰白三烯。CysLTs参与哮踹、过敏性皮炎、心血管疾病、癌症、脑损伤及脑重构等过程。

半胱氨酰白三烯对脑的致损伤作用

目前认为 , 各种脑损伤 ( 脑缺氧、脑外伤 ) 的继发性损伤或称二次脑损伤 (secondary injury) 因素是造成脑损害发生、发展的重要原因。

这些因素涉及:脑缺氧、钙离子超载、氧自由基堆积、炎性因子的刺激等。其中炎症应答 (inflammatory response) 是脑损伤病理生理过程中的一个重要组成环节,半胱氨酰白三烯 (cysteinyl LTs) 包括LTC4, LTD4 和 LTE4 是一类重要的炎症反应介质 , 是花生四烯酸 5-脂氧合酶 (5-lipoxygenase,5-LOX) 的代谢产物。 CysLTs 通过激活半胱氨酰白三烯受体 ( 包括 CysLT1 和 CysLT2 受体 ), 参与多种炎症病理过程 , 介导平滑肌痉挛 (smooth muscle constriction) 、微血管渗漏 (vascular leak) 等。近年研究证明 , CysLTs 除在哮喘、过敏性鼻炎等炎症性疾病发病过程中起重要作用外 , 也参与脑卒中、脑外伤、脑肿瘤、癫痫等中枢神经系统疾病的病理生理过程。脑缺血后CysLTs 释放增加 , 5-LOX 抑制剂能减少 CysLTs 的产生,从而保护脑缺血性损伤。
