StressMarq抗体常见问题汇总

1.我购买的抗体浓度是多少?

我们纯化的单克隆和多克隆抗体浓度可以在产品说明书和网站上找到。未纯化的多克隆抗体和腹水单克隆抗体的浓度无法确定,因为血清中含有除了抗体以外的其他很多蛋白。

2.对于不同的应用我该用的抗体稀释度是多少?

我们强烈建议每一个用户根据他们具体的应用需求和组织类型来决定合适的稀释度。建议的起始抗体稀释度可以在产品说明书上找到。 如果我们没有列出某一个具体应用的稀释度,下面是一些常规建议供您参考:  

纯化抗体 血清 腹水

WB 1/1000 (1 µg/ml) 1/1000 1/1000

IHC/ICC 1/500 (2 µg/ml) 1/100 1/100

3.怎样分量处理抗体?

我们强烈建议您将抗体分成一次用量的小份存于几个小瓶中,以避免多次的冷冻解冻过程。所以请先决定一次实验的用量,然后依据用量分几份保存。需要注意的是少于 5-10 µl 量相对会有更严重的蒸发导致浓度改变较大。

4.缓冲液中 PBS 浓度是多少?

抗体缓冲液中PBS 的浓度是 0.01M (10mM).

5.抗体结合的表位序列是什么?

表位(抗原上与抗体结合的部分的序列) 很少有确定的,如果我们的说明书上免疫原一栏没有注明,那就说明我们不提供相关信息。

6.用于生产抗体的免疫原序列是什么?

我们尽可能地在说明书上提供用于生产抗体的免疫原的序列,但通常来说这属于我们公司专利所以我们没法提供这些信息。

7.我使用的抗体停产了,我该怎么办?

8.这个抗体有没有在其他物种或应用中验证过?

我们致力于随时将新验证的物种和应用更新到说明书上。如果您需要的物种或应用没有列在说明书上,请到我们的产品发表文献一栏查看或者与我们联系 (1.250.294.9065 或 info@stressmarq.com)或咨询代理商上海金畔生物。

9.我怎么预测一个抗体是否有可能和一个未验证的物种有交叉反应?

如果您没有找到验证过某物种的抗体,那么您需要做的是最可能会与该物种有交叉反应的抗体。要想知道会不会有交叉反应,您需要用该抗体的免疫原序列与您的目标蛋白做一个氨基酸序列比对。 我们建议使用 Uniprot (http://www.uniprot.org/) 来查找和做序列比对。该应用会告诉您比对结果相似度的百分比。超过 80%的比对相似度 暗示这个抗体对您的目标蛋白可能有较强的交叉反应。StressMarq 不保证某抗体会在未验证的物种有交叉反应。如果您愿意先测试一下,我们提供免费试用样品,详情请参见抗体试用项目。

10.什么是克隆号?

克隆号是制定给某一个单克隆抗体的杂交瘤细胞株的编号。这个编号是抗体制造者赋予该杂交瘤所特有的号码。它与产品批次/批号是不同的,同一个克隆号可以有多种不同批次。

11.什么是分析证书 (CoA)?

分析证书是一种质量管理文件,用于证明该产品达到了说明书上列出的标准,通常包括证明该产品在某些条件下运作的数据。

12.我该怎么选择合适的二抗?

首先您需要的二抗要能够识别一抗的宿主,例如您的一抗是小鼠抗HSP70 抗体,那么您应该选择抗小鼠的二抗 (例如 羊抗小鼠抗体)。然后您再选择用哪一种抗体标记来显现抗体。
