A causal relationship between centrosome defects (either structural or functional) and cancer development, progression, and aggressive clinical behavior has long been suspected, yet the underlying mechanism(s) have remained enigmatic. However, a recent publication in Nature by Godinho et. al.1 offers an explanation for the link between centrosome amplification and cancers. This group demonstrated that centrosome amplification leads to enhanced Rac1 activity, resulting in compromised cell-cell adhesion and increased invasiveness. This study represents an important step forward in the expanding field of centrosome biology associated with human malignancies.
