RIG-I/MDA-5激动剂及相关产品
RIG-I和MDA-5是细胞浆中的RNA解旋酶,它们识别伴随病毒感染产生的胞内双链RNA(dsRNA)分子模式。尽管RIG-I和MDA-5在结构上具有高度相似性,它们却识别完全不同种属的病毒。RIG-I主要参与副粘病毒Paramyxovirus(例如新城疫病毒Newcastle disease virus和仙台病毒Sendai virus)、棒状病毒Rhabdovirus(例如水疱性口炎病毒VSV)、黄热病毒Flavivirus(例如丙肝病毒HCV)和正粘病毒Orthomyxovirus的识别。而MDA-5则主要识别小核糖核酸病毒Picornavirus(例如脑心肌炎病毒ECMV)和poly(I:C),一种病毒双链RNA的类似合成物。尽管RIG-I和MDA-5所识别的配体完全不同,它们所激活的下游信号通路却是一致的。感受到双链RNA之后,它们被接头分子IPS-1(亦称为MAVS,CARDIF,或VISA)招募到线粒体外膜表面,进而激活包括IRF3、IRF7和NF-B在内的一系列转录因子。IRF3和IRF7控制I型干扰素的表达,NF-B则调控其它炎症因子的产生。
